Clayton Lamb was a tall, powerfully built left footer who marked and kicked well and was a ferocious tackler. He commenced his league career with West Adelaide, whom he joined straight from primary school, in 1981, but it took him a few seasons to establish himself. Capable of playing in almost any position on the ground, Lamb spent nine seasons with the Bloods, during which he played 175 games. He was the club's leading goalkicker with 38 goals in 1988, and captained the side the following year.

In 1990 he crossed to Glenelg hoping for the chance to play in a premiership team, but he was hampered by injury. Lamb was a member in 1991 of the Adelaide Crows' inaugural squad but he was again beset by injury and managed just one AFL appearance. Back with Glenelg in 1992, he produced a much better season of football culminating in participation in his first ever senior Grand Final as the Bays took on, but were badly beaten by, Port Adelaide. During the Glenelg phase of his career Clayton Lamb added 87 league games to his career tally. He represented South Australia four times.
